Biotium CD66 (CEA)(C66/261), CF568 conjugate, 0.1mg/mL
This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA positive) from pleural mesotheliomas (rarely or weakly CEA positive). Recognizes a glycoprotein of 44-88 kDa, which is identified as CD28. It is the critical T-cell co-stimulatory receptor which provides to the cell the important second activation signal by binding CD80 and CD86 that are expressed by antigen presenting cells. Besides its co-stimulation role, CD28 functions in preventing T-cells from anergic hyporesponsive state or from undergoing premature apoptotic cell death. CD28 is also expressed on human fetal NK cells and some NK cell lines, whereas on murine NK cells the CD28 expression is much broader.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. Recognizes a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45RO) (4th Leucocyte Typing Workshop: Code No. N31). The epitope recognized by this antibody is sensitive to neuraminidase digestion. This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. CD6 is a type I transmembrane glycoprotein that contains a 24-amino acid signal sequence, three extracellular scavenger receptor cysteine-rich (SRCR) domains, a membrane-spanning domain and a 44-amino acid cytoplasmic domain. The CD6 glycoprotein is tyrosine phosphorylated during TCR-mediated T cell activation. CD6 shows significant homology to CD5. CD6 is present on mature thymocytes, peripheral T cells and a subset of B cells. This MAb is specific to ER alpha and shows minimal cross-reaction with other members of the family. Epitope of this MAb is mapped between aa120-170. ER is an important regulator of growth and differentiation in the mammary gland. Recognizes a protein of 55 kDa, identified as CD25 (Workshop IV; Code A27). CD25 is expressed on activated T- and B-cells and activated monocytes/macrophages. With respect to lymphomas, CD25 is present on malignant cells of Hodgkin's disease, HTLV-1 adult T-cell leukemia, cutaneous T-cell lymphoma, and hair cell leukemia. Increased levels of soluble CD25 are observed in the leukemias/lymphomas and inflammatory/ autoimmune diseases. CD25 molecule alone appears to function as a low affinity receptor and associates with CD122 (IL-2R beta chain, p75) and CD132 (common gamma chain) to form the high affinity IL-2 receptor complex. CD25 antibodies detect three epitope regions, A, B and C. CD10 (also known as Common Acute Lymphocytic Leukemia Antigen (CALLA)) is a type II transmembrane protein. It is a cell surface enzyme with neutral metalloendopeptidase activity, which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt's, and follicular germinal center lymphomas, and on cells from patients with chronic myelocytic leukemia (CML). Recognizes a protein of 210-220 kDa, which is identified as the complement receptor 1 (CR1)/CD35. This MAb is specific for a site in CR1 that is distal from the C3b-binding site, so that it is unable to block CR1 activity. This MAb is highly specific to CR1 and shows no cross-reaction with CR2. The primary function of CR1 is to serve as the cellular receptor for C3b and C4b, the most important components of the complement system leading to clearance of foreign macromolecules. The Knops blood group system is a system of antigens located on this protein. Follicular dendritic cells (FDC) are restricted to the B-cell regions of secondary lymphoid follicles. They are CD21 /CD35 /CD1a-. Recognizes a protein of 75 kDa, identified as γ heavy chain of human immunoglobulins. It reacts with all sub-classes of γ chain of human immunoglobulins. It does not cross-react with α (IgA), μ (IgM), ε (IgE), or δ (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. This MAb is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The most common feature of these malignancies is the restricted expression of a single heavy chain class.
